The Reeves Lab at UCSF studies tumor heterogeneity and its influence on the anti-tumor immune response. We have developed a unique model system to study heterogeneity using the “Confetti” mouse—a mouse containing a cassette which, when activated, permanently, randomly labels cells one of four colors. This permits us to study heterogeneity by labeling multiple clones in a tumor with different colors and following them side-by-side over time. We are currently hiring a tech/junior specialist, who will specifically be involved studying the impact of tumor composition on the immune response. We have found that some tumor cells, when allowed to form a tumor on their own, are highly infiltrated by T cells; while others are an “immune desert” with few infiltrating T cells. We now plan to use this system to determine by what mechanism these tumor cells dictate the level of infiltrating T cells, as well as how a heterogeneous tumor comprised of both types of tumor cells would behave.
