 Project Title:  Understanding how mTORC1 regulates the efflux of essential amino acids from the lysosome
 Project Description:
The mechanistic target of rapamycin complex 1 (mTORC1) kinase is a crucial regulator of cell growth and metabolism. Recently, using a novel method developed in the lab to rapidly purify lysosomes, we found mTORC1 regulates the lysosomal abundance of essential amino acids through a mechanism that involves the amino acid transporter SLC38A9. We are now working to understand how exactly mTORC1 regulates the transport function of SLC38A9. Understanding this process may be of particular value in Ras-driven pancreatic cancers, which not only depend heavily on micropinocytosis to sustain their amino acid levels, and for cell survival and proliferation, but also require SLC38A9 to form tumors in vivo.
The Sabatini lab is looking for a student who is interested in cancer research, metabolism and biochemistry to assist with this project.  This opportunity is offered for the Fall and Spring terms with possibility for continuation during next summer.  The student will learn a variety of techniques including mammalian cell culture, protein purification and various types of biochemical assays.
