Project Title: Search for Extraterrestrial Genomes: An in situ PCR
Detector For Life on Mars Ancestrally Related to Life on Earth
Project Description
Due to significant meteoritic exchange between Earth and Mars, a
reasonable case can be made that life (past or present) on Mars may
be ancestrally related to life on Earth. We are developing a DNA
detector (based on polymerase chain reaction, or PCR) for in situ
analysis on other planets, most immediately, Mars. This instrument
will allow the detection very low levels of any microbial life on
Mars, and will determine its phylogenetic position by analysis of the
DNA sequence of the genes detected in situ.
We are seeking two highly motivated individuals for paid full-time
summer positions to join our team to (1) design, build, and run
experiments with a bench-top microfluidics-based prototype that uses
some of the technologies developed by the Quake lab at Stanford, and
(2) carry out DNA detection experiments using relevant "extreme
environment" terrestrial samples.
Prerequisites or Requirements for the position:
We are looking for individuals with some of the following skills and
interests:
* Mechanical and electronics design
* Bio-lab experience (PCR, gel eletrophoresis, microfluidics)
* MATLAB, microcontrollers, optics, imaging
This position is located in the Simches building at Massachusetts
General Hospital. You would work as part of our MIT/Harvard/MGH/
Industry team, in a great environment, a 5 minute walk from the
Charles/MGH T station.

Microfocal X-ray localization of electrodes during behavioral neurophysiology
experiments
A central approach in neuroscience is to characterize brain regions using
microelectrode recordings from neurons. Our lab carries out such experiments,
recording from neurons in the inferotemporal cortex of awake, behaving
non-human primates performing visual object recognition tasks. Localizing
electrodes is traditionally accomplished using a combination of stereotactic
guidance for electrode placement and histological confirmation of electrolytic
lesions after experiments, but only with resolution of 2-3 mm. We have recently
developed a novel X-ray system with an order of magnitude better accuracy (~100
microns) for localization of electrodes during neurophysiological experiments,
and have begun our first experiments using this system.
We are looking for a UROP to help develop image analysis software. This project
is well suited for a computer-savvy student seeking experience with X-ray and
MRI automated image analysis and systems neuroscience. In particular, it
involves using matlab code to determine the center of mass for each fiducial
visualized with X-rays, and each vitamin E fiducial visualized with
high-resolution anatomical magnetic resonance imaging (MRI). Later phases will
involve co-registration of X-ray data and MRI data, and GUI design.
Contact: If you are interested, please contact Alex Papanastassiou,
ampapa at mit.edu.
Prerequisites: familiarity with Mac and Windows operating systems. Prior
programming experience would be helpful.
