(6 month or longer position) in Genentech Development Sciences for conducting
and overseeing scientific investigations and screening activities to support a
project using stem cells for generation of functional hepatocytes.
Various cell types have been generated in vitro from stem cells via defined and
stepwise differentiation through the recapitulation of appropriate
developmental steps with stage- and lineage-specific modulators (e.g. cytokines
and small molecules). The specific aims of our project are focused on
identifying small molecules that can mimic the signal requirement during direct
differentiation of stem cell into functionally competent and metabolically
active hepatocytes. Utilizing stem cells the co-op candidate will screen
against a focused as well as large chemical library in order to identify small
molecules which cause generation of functionally competent hepatocytes having
an adult hepatocyte phenotype. The application of such a model will be
wide ranging: improved hepatocyte models for drug discovery programs, enhanced
drug metabolism prediction, and optimized tools for mechanistic understanding
of safety liabilities encountered during small molecule drug development.
This position will enable the co-op student to participate in a high impact
project related to stem cell technologies. In addition based on the
research outcomes and individual contribution, the project will provide an
opportunity to participate at a national level meeting and publish the results.
